Design Rationale:

Replacement of amide in fragment 0107 with pyrazol to additionally addresse carbonyl of Glu166. The nitrile derivative is available from Enamine. Compound with second pyridine moiety to address deeper pocket. Compounds were devised in SeeSAR.
