Benzylic CH may scramble? Replace a chiral centre (CH) with a N. We did it at JBF froma Benzodiazepine to a triazepine and got a clin candidate but it's not a simple one compound switch; I think we made 350 in the end to get there! https://doi.org/10.1021/jm070139l
